Why do BTK inhibitors work for CLL/SLL?

BTK inhibitors work by targeting a specific protein called Bruton's tyrosine kinase (BTK) that helps CLL/SLL cancer cells survive and grow. In CLL/SLL, the body produces malignant B cells (a type of white blood cell), and BTK sends chemical signals that trigger the production of new B cells.

By blocking these signals, BTK inhibitors prevent new CLL/SLL tumor cells from forming and cause existing cancer cells to die off. This helps slow or stop the progression of the disease.

There are different types of BTK inhibitors available:

- Ibrutinib (Imbruvica)
- Acalabrutinib (Calquence)
- Zanubrutinib (Brukinsa)
- Pirtobrutinib (Jaypirca)

Some BTK inhibitors bind permanently to the BTK protein (like ibrutinib), while newer ones like pirtobrutinib bind reversibly and can still work even if mutations develop. This is why BTK inhibitors have become a first-line treatment option for many people with CLL/SLL.
